Girl Talk is one of those things you either love or hate. I choose love friends.
For those of you who have no idea who/what Girl Talk is, he's the crown prince of mashup. What makes him special is mostly an issue of sheer quantity; he often stuffs 20 or 30 samples, often explicit enough where the careful listener can identify them, into a 3 minute song. It's very different and very fredgy.
In addition to just sitting back with a good pair of headphones and trying to pick out the samples, I'd also suggest listening to it with the wiki (which lists all of the identified samples) and trying to figure out what is what. Alternatively, you can watch these bizarre videos, which mash up the music videos of the sampled songs in a fashion equally disorienting and strangely appealing to the music itself.
Lastly, you can just watch this video, for one of the best songs of 2007, which is liberally sampled from in the first song on the new Girl Talk record:
